Type 2 Diabetes Mellitus:
As an initial therapy alongside diet and exercise to improve glycemic control.
As a second-line therapy when diet, exercise, and initial treatment with a sulfonylurea or metformin alone do not result in adequate glycemic control.
Dosage:
Starting Dose: 250 mg/1.25 mg once or twice daily with meals.
Maximum Dose: 2000 mg of metformin and 20 mg of glibenclamide per day.
Administration: Take with meals to reduce gastrointestinal side effects and to minimize the risk of hypoglycemia.
Titration: Increase the dose gradually based on blood glucose response and tolerability.
Precautions:
Not recommended for patients with severe renal impairment, lactic acidosis, or ketoacidosis.
Use with caution in patients with liver disease.
Regular monitoring of blood glucose and renal function is advised.
Avoid alcohol consumption as it may increase the risk of lactic acidosis.
